研究室概述:
循环糸统对组织血流的调节主要通过血管的收缩与舒张实现,我们室的主要研究邻域为生理与病生理条件下血管反应的调节机制,特别是一氧化氮-鸟苷酸环化酶-环鸟苷酸蛋白激酶,RhoA与RhoA蛋白激酶,肌球蛋白磷酸酶,活性氧,与活性氮等ROCK对血管的收缩与舒张的调节机制。我们近年来的工作表明环鸟苷酸蛋白激酶在硝酸甘油与一氧化氮引起的冠状动脉舒张起关键作用,环鸟苷酸蛋白激酶的下调是硝酸甘油耐受产生的机制之一。我们的工作还表明肌球蛋白磷酸酶亮氨酸拉链表达阳性亚单位的蛋白酶体降解增加可导致冠状动脉与肺动脉对一氧化氮舒张减弱。
Research Description:
Vasoconstriction and vasodilatation are the major means by which the blood flow to the tissues is regulated. Our research interest is primarily in the mechanisms underlying the vasoreactivity under physiological and pathophysiological conditions, in particular, the role of nitric oxide (NO), soluble guanylyl cyclase (sGC), cGMP-dependent protein kinase (PKG), RhoA/Rho kinase, myosin light chain phosphatase (MLCP), and reactive oxygen/nitrogen species. Our recent studies demonstrate that PKG plays a major role in relaxation of coronary arteries and veins induced by nitroglycerin and NO. The downregulation of PKG expression and activity contributes to the development of nitrate tolerance. Our studies also show that an enhanced degradation by proteasomes of the leucine-zipper positive isoform of the regulatory subunit of MLCP may lead to diminished response of coronary and pulmonary arteries to nitric oxide.
